I'm determined to enter my R.O.B. amiibo in future tournaments, can anyone tell me if I should change his setup?
60/60/0 -Critical Hit -Improved Escape -Lifesteal
CUSTOMS: High-Speed Burner, Slip Gyro
Note: I've tried Overloaded on R.O.B. It didn't work out, so I'm trying my hardest to avoid negative Speed.

I think the spread is overall good. I can't really say much as I don't have experience with R.O.B. (as an amiibo nor playing as him).
Though here's a thought: since negative speed didn't work out, have you tried adding a bit of speed? Something like 50/60/10 or 40/60/20.

I considered adding speed one time, but since his recovery is already great, I decided against it. I wanted to focus on the offensive and defensive aspects of him. Now that I look at it, the spread IS good, but I may be considering changing his bonuses.

Attack points are especially important for R.O.B., since his damage output is very lacking, despite being a heavyweight. That, or you focus really hard on defense. Because without that firepower, R.O.B. will be tackling on damage very, very, slowly lol
Also, giving R.O.B. speed points wouldn't be a bad thing. If he gets knocked into the air, he's very well open to an up smash on his way down because of his extremely poor air speed. If you're against a 200/120/-200 Ganondorf though, I wouldn't be too concerned about it.
If you're looking for R.O.B. to use his down special, I'd definitely recommend you give him his Flaming Gyro custom (or whatever it's called...). R.O.B. refuses to use the normal Gyro because of it being so unsafe, but he'll use custom Gyros because of their lingering hitboxes.
So, Lifesteal, simply put, stinks on R.O.B. That bonus really only benefits characters with the most damaging moves or characters that have extremely high hit-consistency. In this case, R.O.B. doesn't really fit the bill for either lol I'd recommend Auto Heal over Lifesteal because of this.
Speaking of bonuses, Explosive Perfect Shield is amazing with R.O.B. because of the wide radius it covers.
Y'know what, actually, just give him the same bonuses as your Luigi, it would work wonders for R.O.B. now that I think about it lol

Okay... I just changed his stats to 100/100/-80 AND I Square One'd him to Level 1. I temporarily changed his bonuses to Improved Shield Regeneration, Mirror Shield, and Health-Restoring Shield; I'll change these to match my Luigi's once I feel R.O.B.'s got the hang of relying on his shield.
